GAM-05, a first-in-class mitochondria-targeted Hsp inhibitor, is advancing as a novel oncology candidate with strong potential in combination regimens. Preclinical data demonstrate synergistic activity when paired with marketed chemotherapies such as doxorubicin, platinum agents and taxanes.
GAM-05’s unique mitochondrial mechanism offers complementary benefit alongside targeted therapies currently in development, including PARP and kinase inhibitors, with the potential to overcome resistance pathways.
The in vivo imaging data demonstrate that combining GAM-05 with BEZ produces substantially greater suppression of glioblastoma burden than either monotherapy. While GAM-05 and BEZ alone each reduce tumor signal relative to vehicle, the BEZ + GAM-05 combination yields the lowest residual signal across animals, consistent with a synergistic or strongly additive antitumor effect.
